Reduction in Fatigue Symptoms Following the Administration of Nutritional Supplements in Patients with Multiple Sclerosis

Abstract

Despite recent advances in immune-modulatory drugs, pharmacological therapies have been proven ineffective in severe presentations of multiple sclerosis (MS), including secondary progressive MS. At present, therapeutic interventions’ performance is primarily focused on ameliorating symptoms to improve the patient’s quality of life (QOL). Among complementary treatments, nutrition has been considered a decisive factor to control symptoms and enhance the wellness of MS patients. Although no special diets are associated with MS, the impact of diet and dietary supplements on the course of progressive forms of the disease has been studied during the last few years. Fatigue is among the most common and disabling symptoms reported by MS patients. Fatigue has been defined in the Multiple Sclerosis Council for Clinical Practice Guidelines (MSCCPG, 1998) as a “subjective lack of physical and/or mental energy that the individual perceives as an interference with habitual and desired activities”. This study aimed to compare the psychometric functioning of the “Fatigue Severity Scale” (FSS) and the “Modified Fatigue Impact Scale” (MFIS) in our sample of people with MS. Specifically, during chronic treatment, the change in these two parameters with two vitamin-rich dietary supplements (Citozym® and Ergozym®) was evaluated. The impact of these nutritional supplements revealed differences in antioxidant and anti-inflammatory parameters among the volunteers in the treatment group, with a subsequent improvement in fatigue. In conclusion, the results obtained have confirmed the effectiveness of complementary nutritional therapies, evaluated essentially based on hematological biomarkers, through which it is possible to act on disability to improve the QOL of MS patients.

Abstract

Weight loss in patients with cancer is caused by cancer cachexia and chemotherapy-induced nausea and vomiting. Recent developments in antiemetic drugs have substantially improved nausea and vomiting, but this intervention did not reduce weight loss and other more severe side effects of chemotherapy, like anorexia, weakness, cough, dyspnea, hemoptysis, and pain. This study aimed to investigate the effects of nutrition intervention with a food supplement, during chemotherapy in patients with advanced nonsquamous non-small cell lung cancer (NSCLC). Patients received individualized nutrition counseling by a registered dietitian and were provided with oral supplements of Texidrofolico® for 90 days. Bodyweight and the mentioned other side effects were evaluated at baseline and after 90 days of intervention. To assess the effects of this dietary supplement, a total of 30 patients were retrospectively enrolled as controls, and the bodyweight and change in side effects of chemotherapy were compared with those observed in 30 Texidrofolico®-treated patients. After 90-day intervention, by oral supplement of Texidrofolico®, the patients, during the course of cytotoxic chemotherapy, showed an improved quality of life and not significant weight and BMI loss respect the control group. Furthermore, the number of patients, treated with Texidrofolico® who maintained or increased their body weight, after 90 days of treatment was significantly higher than in the control group. The effects of treatment with the food supplement have also been studied from a metabolic point of view. It was possible to find that one of the known markers of tumor growth, plasma polyamines, was reduced after the treatment. A possible relationship between these biogenic amines and the folate cycle is discussed. In conclusion, early intensive nutrition intervention with oral supplements of Texidrofolico® during chemotherapy of NSCLC patients prevents weight loss and it is beneficial for their quality of life.

Regression of Carotid Plaques in Individuals at Low-to-intermediate Cardiovascular Risk Treated with Citozym and Propulzym

Abstract
A prospective trial was performed to assess coronary plaque regression following about 5 months of treatments with two dietary supplements, Citozym and Propulzym. Coronary segments that included the most diseased plaque of 20 selected patients were analyzed using carotid ultrasound (CDUS). Inclusion criteria were as follows: 20 subjects (10 of control group and 10 of test group) aged between 54 and 74 years with a clear diagnosis of carotid plaque and stenosis percentage of not less than 40% and not more than 70%. The data obtained showed a wide inter-individual variability, with a range of reduction from 4% to 27%. It is interesting to note that the plaque reduction in one patient reached a value of about 27% observable with the ultrasonic examination. Since it is clear that carotid plaque morphology/stability is one of the most important factor regarding the definition of cardiovascular risk, our preliminary results suggest the possibility of a non-invasive treatment of carotid plaque.

Preventive Effects of A Mixture of Micronutrients with Antioxida-tive Properties on Experimentally Induced Prostate Hyperplasia

Abstract
A mixture of antioxidants found in a commercial food supplement called Citozym (CIZ), showed a preventive capability on experimental prostate hyperplasia induced by testosterone propionate (TP) in C57BL6/N mice. Treatments by CIZ were given orally and started 10 days before the induction of prostate hyperplasia. TP significantly increased prostate size and weight ratio, and this induced increase was significantly inhibited in CIZ-treated mice in comparison with positive controls in a dose-dependent manner. We conclude that CIZ can prevent TP-induced prostate hyperplasia and therefore may be beneficial in the management of benign prostatic hyperplasia.

The Influence of Preventive Multiple Micronutrients Supplementation on Liver Steatosis in High-cholesterol Fed C57BL6/N Mice

Abstract
Liver steatosis development was obtained after high cholesterol diet (HCD) in C57BL6/N mice. Mice were preliminarily treated for 10 days with multiple micronutrients contained in a commercial food supplement called Citozym (CIZ), and successively for 40 days with CIZ and HCD, to compare the protection effect of used compounds on liver metabolism and metabolic steatosis. At the end of the experiments, livers were dissected for histological examinations. Plasma total cholesterol (TCH), triacylglycerol (TAG) aspartate aminotransferase (AST), and alanine aminotransferase (ALT) concentrations were significantly higher in all HCD-fed mice. These data support a lipotoxic model of cholesterol-mediated hepatic steatosis. We found that the administration of CIZ to HCD-mouse model of steatosis significantly decreased plasma TCH, TAG, ALT and AST levels along with a reduction of the accumulated fat and inflammation in the liver. These findings suggest that a preventive and continuative treatment of CIZ exerts a protective effect against metabolic hepatosteatosis.

Multiple Sclerosis: Effect of Oral Administration of an Antioxidant Dietary Supplement in C57BL6/N Induced Model of Experimental Autoimmune Encephalomyelitis

Abstract
In the pathogenesis of demyelinating diseases including multiple sclerosis (MS) an important role is played by oxidative stress. Increased energy requirements during remyelination of axons and mitochondria failure is one of the causes of axonal degeneration and disability in MS. In the presence of neurological diseases such as MS, F2-isoprostanes are concentrated in higher quantities. In this context, we analyzed the levels of F2-isoprostanes in the plasma and cerebrospinal fluid of mice of the strain C57BL6/N with an induced experimental autoimmune encephalomyelitis (EAE), orally treated with two concentrations of Citozym, a dietary supplement with evident antioxidant properties. Compared to the control group, Citozym-treated EAE-mouse had significantly lower levels of F2-isoprostanes both in plasma and in cerebrospinal fluid. Furthermore, according to the guidelines of IACUC, treatment with Citozym at higher concentrations drastically reduced neurological signs of induced EAE.

Reduction of Breath Alcohol Levels in Healthy Subjects by Citoethyl

Abstract
The level of alcohol in the blood is a problem not only for health but also for motorists during the driving. The search for dietary supplements capable of reducing the rate of alcohol was hectic and different products with dubious quality have been put on the market. In the present work, 60 healthy male individuals were subjected to two tests. The first included the administration of 15 mL of concentrated commercial mixture of vitamins and antioxidants called Citoethyl (CTH), 40 minutes prior drinking 80 mL of pure ethyl alcohol diluted in 200 mL of water. The second test was performed by administering 15 mL of concentrated CTH, 40 minutes after drinking 80 mL of pure ethyl alcohol diluted in 200 mL of water. The amount of alcohol ingested corresponded to 1600 mL of beer ( 5% v/v alcohol) or 535 mL of wine (15% v / v alcohol) or 200 mL of liquor (40% v / v alcohol). The breath alcohol test was performed on each individual in six ranges of time from the administration of alcohol. We report in this work that there is a net reduction of the breath test value for alcohol resulting from the intake of CTH, in all the conditions used for experimentation. Not guaranteeing the results presented for higher doses of alcohol and for a too short period of time of administration of the CTH. The present work does not pretend to suggest a product for making drunk driving safer, but simply reports the experimental data on a mixture of vitamins and antioxidants, that show its qualities in reducing the rate of breath alcohol levels in healthy subjects.

Organ Culture Model of Liver for the Study of Cancer Treatment for Hepatocellular Carcinoma

Abstract
The liver, the largest organ of the human body, is a multifunctional organ with various metabolic activities that plays a fundamental role in maintaining the body and in sustaining life. Although the liver has great regenerative capacity and recovery, the damage caused by chronic diseases such as cancer or viral infections can lead to permanent loss of liver function. Studies on the mechanism of liver disease, have focused on the selection of cell and tissue culture techniques, including strategies based on in vitro models. The organ culture is a promising tool for the study of liver diseases, because it can mimic the complex of the microenvironment in vivo using a three-dimensional model of human liver tissue. These models allow a better study of the specific functions of the liver. In this context, we have analyzed the development of a hepatocarcinoma, obtained by inoculating a murine hepatocarcinoma cell line, Hepa 1/A1s, in the liver of 10 mice of the strain C57BL / 6. After 20 days from the inoculation, the portion of liver invaded by the tumor was removed from the animals and cultured. A group of 5 liver explants were used as a control and other 5 explants were cultured for 4 weeks in a complete medium containing 10% Citozym, a food supplement with reported antioxidant properties. The cancer-invaded hepatic lobes, treated with Citozym, showed a clear reduction of the weight and the volume of the hepatic tumors, when compared with the control explants.

Correlations Between Administration of Food Supplements with Marked Antioxidant Properties and Clinical Parameters in Patients with Prostate Carcinoma

Abstract
Several studies indicate that high oxidative stress is associated with various degenerative diseases, including tumors. The high levels of free radicals present in many patients derive from the chronic lack of antioxidants, caused by an increasingly poor and artificial diet. The study presented in this research, conducted on 50 male volunteers, carriers of Prostatic Carcinoma (PCa) at different stages of development and under current therapy, showed that the daily intake of antioxidants contained in two food supplements, Citozym (CIZ) and Propulzym (PRZ), induces a significant increase of the ratio free/total PSA and also a reduction of various clinical parameters, correlated with PCa. This result suggests a potential slowdown in the progression of the disease. This study was planned on the basis of a recent preliminary trial, that highlighted the positive activity of CIZ in a model of benign prostatic hypertrophy. Studies are in progress to identify the components present in these food supplements affecting biochemical signals, elements that underlie the ability of PCa to progress. This does not mean that the intake of antioxidants is a cure for PCa, but that a high undiagnosed oxidative stress, much less correct by the standard oncological approaches, contributes to the evolution of the tumor disease. In other words, current oncology focuses correctly on reducing the tumor mass, but does not intervene on the biological medium that produced it. This is why there is still a dramatic incidence of relapses.

Effect of a Novel Dietary Supplement on Cancer Cells Metabolism

Abstract
One area of research currently garnering significant attention is the in-depth study of cancer cell metabolism. The purpose of this research is not exclusively to discover new anticancer therapies, but rather to improve the quality of life of the patient. Many cancer patients suffer from a muscle wasting syndrome called cachexia. Cancer related cachexia impairs quality of life and response to therapy, which increases morbidity and mortality among cancer patients. The data presented in this report were collected to address disorders caused by therapies aimed at reducing tumor growth. Carbohydrate metabolism is the major pathway in the cell providing energy and building blocks for macromolecule biosynthesis. Glucose metabolism in cancerous cells is remarkably different from that in their normal counterparts. The purpose of this research was to investigate the effects on energy metabolism in normal and neoplastic cells of the various components of the dietary supplement Texidrofolico (TXF), which act on their cellular enzymatic complexes. Treatment of HepG2 tumor cells with TXF reduces glucose consumption, and slows reduction of pyruvate to lactate, following indirect inhibition of lactate dehydrogenase (LDH) by blocking regeneration of NAD+ from NADH. These activities inhibit glycolysis, with consequent reduction in energy yield, evidenced by an increased ADP/ATP ratio. By contrast, in normal MEF cells treated with TXF, the ADP/ATP ratio was observed to be lower than in untreated control cells, showing increased energy relative to untreated normal cells. The observed effects seem to be due to the presence of two active components in TXF: folic acid, and pyruvate. Calciferol, which exerts less influence on glucose consumption, instead shows some inhibition of LDH reductase activity.

Effect of Oral Administration of Dietary Antioxidant Supplements in Patients with Chronic Hepatitis C

Abstract
In a prospective, randomized and single-blinded clinical trial, we compared patients with Chronic hepatitis C (CHC) orally treated with sucrose diluted with water (1:2) twice a day for 56 days (control group), with patients orally treated with four antioxidant dietary supplements rich in vitamin B5, B9, C, D, citric, pyruvic, and tartaric acids and carbohydrates (CCEP: Citexivir, Citozym, Ergozym Plus and Propulzym). The efficacy of treatment was evaluated once a week for 8 weeks, by monitoring changes in the activities of circulating Alanine aminotransferase (ALT), Aspartate aminotransferase (AST) and Gamma-glutamyl transferase (GGT) as markers of liver damage. After a treatment of 84 days the viral title was evaluated through the HCV-RNA compared with the levels of anti-C100-3. The security and tolerability of the treatment were evaluated on the basis of clinical adverse events and results of laboratory tests. The experimental data obtained showed that the oral treatment of patients suffering from HCV infection of genotype 1, with CCEP, markedly influenced the values of the three enzymatic markers of hepatic disease. The data presented also showed the reduction of viral replication evidenced by the rate of HCV-RNA levels. As reported by others, we confirmed the low reliability of the research of anti-C100-3. This research is not meant to suggest the treatment reported as a therapy for the treatment of HCV infection, but data obtained may tend towards the possibility of administration of a dietary supplement such as CCEP in support of the official drug therapy of CHC in the nutritional care of HCV patients.

Administration of the chemotherapeutic agent by organ perfusion is more effective than the systemic administration. The active drug is reached by the tumor, to a higher concentration, and normal tissues are spared from major damages. The perfusion of various areas of the body is a recent goal of the experimental and clinical trials. Fatal malignancies observed in humans occur mainly in the abdominal area of the body, The development of a clinically applicable technique for the perfusion of this area is desirable. This report covers the laboratory studies, carried out in the development of a liver perfusion technique for chemotherapy. The results obtained have allowed us to highlight that, the potential anticancer activity of Citozym, a micronutrients supplement, is likely the result of the inhibition of oxidative stress. This work is the preliminary evidence, of the possible mechanism by which Citozym exerts its antineoplastic activity.
